Our partner RUG have published a detailed protocol to generate mutant libraries experimentally by primer design, mutagenesis, protein production and screening and then mutation combination strategies that can yield stabilized mutants with increases in apparent melting temperature of 20–35°C and vastly increased half-lives with resistance to cosolvents. These mutants were designed using algorithms to predict the differences in folding energy of a mutant compared to the wild-type using the highly successful FRESCO workflow method (A Computational Library Design Protocol for Rapid Improvement of Protein Stability: FRESCO).
